The runaway World No.1 is looking for back-to-back victories in Ohio.
Scheffler, 28, stormed to the top of the leaderboard by picking up four birdies in his final five holes on Saturday evening.
His 4-under 68 gave him a one-shot lead heading into the final round of the penultimate signature event of the 2025 campaign.
It means he is searching for his third victory in his last four starts having previously won the second men's major of the year - the PGA Championship - and the Charles Schwab Challenge.
If Scheffler wins, it would take him to 16 PGA Tour titles - including three major wins and an Olympic gold medal - since his breakout victory in February 2022.
Scheffler has also set up a golden chance to become the first player to successfully defend the Memorial Tournament since Tiger Woods in 2001.
Woods won three Memorial Tournaments in a row from 1999.
Ominously for the likes of Griffin and the chasing pack, Scheffler has also converted all eight of his previous 54-hole leads.
Griffin, who is looking to win his second PGA Tour title in 14 days, told reporters that every player wants to compete and ultimately prevail against Scheffler.
"I would like to say that's what all of us out here on Tour wish for," he said.
"They want to compete against the best players. I definitely do. I want to stack up my game against his. I feel like right now -- you know, I beat him last week.
"Obviously, he's coming off a major win. But, yeah, I feel like he obviously can be beaten, and I've just got to keep the pedal down and make a lot of birdies because I know he's going to as well."
Griffin added: "But I've got to do some similar things to what I did on Thursday when I had an awesome round.
"Obviously, I made some of those putts that you don't normally make, and so if you look at the last couple days, I've still hit it really well, but I haven't quite made as many of those longer putts that I did the first day, so hopefully I can sneak a few of those in."
Canada's Nick Taylor is also in the hunt and is three strokes behind Scheffler.
Also in the chasing pack are Keegan Bradley, Jordan Spieth, Sepp Straka and Shane Lowry.
